I am thinking about using 40mm x 40mm bases with four infantry figures on them, in 2 ranks of 2. Will this be compatible with most rules sets where each unit represents a regiment? Thanks.

I've been tempted to do the same; I think it is a safe stand size.

It would work fine. I've seen that scheme several times before and it is flexible and looks good.

And frankly almost any rule set will work with any basing with only minor tweaks. Just so long as both sides are based the same…

That is what I have done with my Perry 28mm ACW's used for Black Powder.

Well apart from the fact mine are actually on 35x35 and I don't us Black Powder! :o)

Seriously though, 4 figures on a square base is the way to go. It makes columns look good and gives two rank line a nice tight appearance.

Speaking of which, that's why I went for 35mm – for my taste 40 leaves too much daylight between figures.
